Humanized Mediator Release Assay as a Read-Out for Allergen Potency
نویسندگان
چکیده
Mediator release assays analyze in vitro immunoglobulin E (IgE)-mediated degranulation and secretion of mediators by effector cells, such as mast cells basophils, upon stimulation with serial dilutions putative allergens. Therefore, these represent an essential tool that mimics the vivo process, which occurs allergen exposure sensitized patients or skin prick tests. Additionally, are usually employed to investigate allergenic potential proteins reactivity patients' sera's reactivity. Herein, we describe a simple 2-day protocol using immortalized rat basophil leukemia cell line transfected humanized human high-affinity IgE plasma-membrane receptor (FcεRI). This variant mediator assay is robust, sensitive, reproducible cell-based system without need immobilize antigen solid matrices. The consists following steps: (1) complement inactivation sera, (2) harvesting, seeding, passive sensitization (3) cause release, (4) measuring β-hexosaminidase activity surrogate for released inflammatory mediators, histamine. represents useful assess capacity allergen-IgE cross-linking trigger can be implemented standardize extracts, compare minor major allergens extracts (pollen, cat dander, etc.), potency homologs, isoforms, fold-variants (e.g., hypoallergenicity), well effects ligands on activity. A more recent application includes use monitor treatment efficacy course immunotherapy.

A Dual Read-Out Assay to Evaluate the Potency of Compounds Active against Mycobacterium tuberculosis
Tuberculosis is a serious global health problem caused by the bacterium Mycobacterium tuberculosis. There is an urgent need for discovery and development of new treatments, but this can only be accomplished through rapid and reproducible M. tuberculosis assays designed to identify potent inhibitors. We developed an automated 96-well assay utilizing a recombinant strain of M. tuberculosis expres...

The development of reliable and clinically relevant potency assays is essential to the practice of safe and effective allergen-specific immunotherapy. Allergen standardization in the United States is based on the establishment of a national reference assigned with a biological potency unit to which manufacturers' products are compared using validated relative potency assays. This ensures, at le...
متن کاملMultiplex Assay for Protein Profiling and Potency Measurement of German Cockroach Allergen Extracts.
BACKGROUND German cockroach (GCr) allergens induce IgE responses and may cause asthma. Commercial GCr allergen extracts are variable and existing assays may not be appropriate for determining extract composition and potency. OBJECTIVE Our aim was to develop a multiplex antibody/bead-based assay for assessment of GCr allergen extracts. METHODS Single chain fragment variable (scFv) antibodies...

A potency assay for Bordetella bronchiseptica bacterins has been developed using mice. The immunogenicities of three bacterins, B, C, and D, were evaluated for ability to prevent death in mice as compared with a reference standard bacterin (RSB-A). Bacterins RSB-A, B, and C were evaluated for ability to prevent death in mice as compared with a reference standard bacterin (RSB-A). Bacterins RSB-...

The aim of this study was to develop a novel model for studies of mediator mechanisms involved in the late asthmatic reaction in the lower airways, by using the sensitized pig. The release of histamine and cysteinyl-containing leukotrienes (cys-LTs), as well as the levels of inflammatory cells in blood and bronchoalveolar lavage fluid, were determined and their relationship to plasma cortisol l...
